Clinical significance of high focal adhesion kinase gene copy number and overexpression in invasive breast cancer

摘要: Increased expression and activation of focal adhesion kinase (FAK) was reported to be an unfavorable factor in various human cancers, including breast cancer. In order study FAK gene status as a prognostic factor, we evaluated copy number protein invasive Tumors from 435 patients with cancer were for using fluorescence situ hybridization (FISH) immunohistochemistry (IHC) based on tissue array method. Survival analyses performed the Kaplan–Meier method 267 patients. 42 out 362 evaluable cases (11.6%) showed high polysomy 22 (6.1%) had amplification by FISH. 108 393 (27.5%) overexpression IHC. FISH positivity significantly associated higher histologic grade, T stage, negative estrogen receptor expression, progesterone triple-negative phenotype; grade phenotype. noted 57.8% (37 64) FISH+ cases. The concordance IHC results observed 74.9% (271 362). revealed shorter overall survival relapse-free compared those FISH− node-positive who received postoperative systemic treatment, 5-year rates.
