Application of Precipitating Activity of Canavalia lineata DC Lectin for Isolation of α2-Macroglobulin from Human Plasma

摘要: To isolate α2-macroglobulin from human plasma, complex of Canavalia lineata DC lectin and plasma was used as a starting material. The solubilized with 0.08 M phosphate buffer pH 7.2 containing 0.9% NaCl 0.02 α-methyl-D-mannoside. solution applied onto Bio gel A-5m column eluted solubilizing buffer. Of three peaks, only the second peak contained α2macroglobulin. After equilibrating 0.06 potassium 6.8, fraction hydroxylapatite same eluate confirmed to be extremely pure α2-macroglobulin; immunoelectrophoresis anti-human serum produced single precipitation line, SDS disc electropho-resis gave band at α-globulin region. Few differences in carbohydrate composition between this preparation those prepared by other methods were observed. By method, about 45% armacroglobulin present recovered.
