摘要: Recent studies describing the mutational landscape of head and neck squamous cell carcinoma (HNSCC) on a genomic scale by our group others, including The Cancer Genome Atlas, have provided unprecedented perspective for understanding molecular pathogenesis HNSCC progression response to treatment. These confirmed that mutations TP53 tumor suppressor gene were most frequent all somatic alterations in HNSCC, alluding importance suppressing development this disease. Clinically, are significantly associated with short survival time resistance radiotherapy chemotherapy patients, which makes mutation status potentially useful factor risk stratification predictor clinical these patients. In addition loss wild-type p53 function dominant-negative effect remaining p53, some mutants often gain oncogenic functions promote tumorigenesis progression. Different may possess different gain-of-function properties. Herein, we review up-to-date information about available via Atlas-based analysis discuss current potential tumor-suppressive role focusing activities mutations. We also summarize knowledge regarding use as evaluation or biomarker prognosis Finally, possible strategies targeting HNSCCs bearing J. Cell.
