摘要: A number of monoclonal antibodies (MoAbs) against the 16 kDa antigen Paramphistomum gracile (16 kDaAgPg) were produced in vitro by hybridoma technique. Reactivity and specificity these MoAbs evaluated ELISA immunoblotting assays. Seven MoAb clones selected from stable clones, namely 1D10, 2D7, 3B10, 3D9, 4F1, 4G4, 5G12. It was found to be IgM kappa light chain isotypes. By ELISA, all reacted with purified 16 kDaAgPg at molecular weight (MW) native MW whole body (WB) excretory-secretory (ES) fractions, but not tegumental antigens (TA) adult fluke. All showed no cross-reactions other parasites commonly ruminants, including Eurytrema pancreaticum, Gigantocotyle explanatum, Schistosoma spindale, Moniezia benedeni, Avitellina centripunctata, Haemonchus placei, Trichuris sp., Setaria labiato-papillosa. Localization distribution 16 kDaAg P. immunohistochemistry, using as probes, that present high concentration cytoplasm vitelline cells, eggshell globules, shells eggs, tegument, muscle, parenchymal cecum This finding indicated is a copiously expressed parasite protein released into ES; thus, its could good candidate for immunodiagnosis paramphistomosis ruminants.
