摘要: We developed versatile low-cost arrays of sol-gel-encapsulated enzymes (referred to as solzymes) suitable for repeated assays bioactivity or enzyme inhibition. Sol-gel microstructures containing active were stabilized on glass at moderate pH and room temperature without harsh calcination. A multi-well bilayer polydimethylsiloxane was used support the solzyme array contain reaction medium. Each 147 microwells has a working volume 5 μL contains 50 μg immobilized enzyme. The maintained high activity through applications exhibited superior thermostability compared soluble enzymes. Among lipases, glucose oxidase, horseradish peroxidase. Twenty different lipases proteases also prepare hydrolase array, which bromthymol blue served generic indicator activity. relative activities encapsulated hydrolases correlated closely with those hydrolases, illustrating that sol-gel encapsulation preserved hierarchy development paves way higher throughput screening diverse proteins enzymes, including are available only in trace amounts. © 2002 Wiley Periodicals, Inc.
