摘要: Several genome engineering applications of CRISPR-Cas9, an RNA-guided DNA endonuclease, require precision control Cas9 activity over dosage, timing, and targeted site in organism. While some dose time have been achieved using small molecules, spatial light, no singular system with all the three attributes exists. Furthermore, reported small-molecule systems lack wide dynamic range, background absence controller, are not biologically inert, while optogenetic prolonged exposure to high-intensity light. We previously a small-molecule-controlled dosage temporal control. By photocaging this activator render it inert photoactivatable, employing next-generation protein approaches, we built low background, fast photoactivation low-intensity light rendering inert. anticipate these controls will propel development practical Cas9.
