Rapid diagnosis of the infection of pine tree with pine wood nematode (Bursaphelenchus xylophilus) by use of host-tree volatiles.

摘要: Attraction of the Bursaphelenchus xylophilus nematode toward 18 volatiles Pinus species was evaluated by a Petri-dish bioassay under laboratory conditions to develop rapid diagnostic kit. Among these compounds, α-pinene, β-pinene, and camphor showed significantly higher attractiveness B. in both reproductive dispersal stages, whereas compounds were not active against mucronatus. A trap tube developed as kit, which consisted filled with 0.8% agar matrix impregnated an attractant: or camphor. All tested attracted number than that control treatment. No significant difference observed among attractants. The cotton-ball more effective filter-paper for attracting artificial pupal chamber bioassay. In pine wood (PWN)-infected tree logs, initially after 8 h period increased time. using most xylophilus. semiochemical-based tube-trapping method is simple use, requires minimal labor, economical detecting living host trees during field sampling.
