Effects of 23-epi-26-deoxyactein on adipogenesis in 3T3-L1 preadipocytes and diet-induced obesity in C57BL/6 mice

摘要: Abstract Background The ethanolic extract of Actaea racemosa L. (Cimicifuga (L.) Nutt.) has recently been reported to ameliorate obesity-related insulin resistance, hyperlipidemia, and fatty liver in rodents. However, it remains unclear which A. components are responsible for these beneficial effects. Purpose We aimed examine the anti-obesity potential 23-epi-26-deoxyactein (DA), is contained extracts racemosa. Study design methods To evaluate effects DA on adipogenesis 3T3-L1 preadipocytes diet-induced obesity C57BL/6 mice, vitro vivo tests were performed. For assessment, we used Oil red O staining that showed lipid accumulation differentiated cells. tests, male 5-week-old mice fed with low-fat diet (LFD), high-fat (HFD), HFD 10 mg/kg/d luteolin (LU; positive control drug), 1 mg/kg/d DA, 5 mg/kg/d 12 weeks, respectively. Glucose tolerance performed at week 17. deposition adipose tissue was visualized by hematoxylin eosin staining. Real-time PCR mRNA levels genes involved adipogenesis, lipogenesis, lipolysis. AMPK signaling SIRT1-FOXO1 pathway assessed real-time western blot. Results 10 μM 20 μM LU treatments inhibited through down-regulating expression C/ebpα, C/ebpβ, Pparγ, critical adipogenic transcription factors. results significantly lowered body weight gain, fat mass, HFD-fed mice. Meanwhile, also reduced resistance serum lipoprotein Mechanistic studies promoted adipocyte lipolysis activating pathway. Conclusion indicate suppresses preadipocytes. treatment ameliorates suggesting a promising natural compound related metabolic diseases.
