摘要: To understand more clearly the link between osteoarthritis and hyperlipidaemia, we investigated inflammatory macrophage subsets macrophage-regulated matrix metalloprotease-3 (MMP-3) A disintegrin metalloprotease with thrombospondin motifs-4 (ADAMTS4) in synovial (ST) adipose tissues (AT) of osteoarthritic mice hyperlipidaemia (STR/Ort). CD11c(+) F4/80(+) CD11b(+) populations ST AT 9-month-old STR/Ort C57BL/6J were characterized compared by flow cytometry real-time polymerase chain reaction (PCR) analyses. Expression tumour necrosis factor (TNF)-α, MMP-3 ADAMTS4, response these factors to anionic liposomal clodronate induced-macrophage depletion also evaluated PCR. TNF-α cells, which isolated magnetic beads, was CD11c(-) cells. In addition, effect on cultured fibroblasts adipocytes investigated. macrophages increased mice. The cell fraction highly expressed TNF-α. MMP3 AT, decreased upon depletion. treatment markedly up-regulated MMP-3. identified as a common subset hyperlipidaemia. induction inflammation may be part mechanism that regulates expression through Our findings suggest numbers elevated levels explain relationship OA.
