摘要: CD73, ecto-5'-nucleotidase, is the key enzyme catalyzing conversion of extracellular AMP to adenosine that controls vascular permeability and immunosuppression. Also prostatic acid phosphatase (PAP) possesses ecto-5'-nucleotidase/AMPase activity present in leukocytes. However, its role related immune system unknown. Therefore, we analyzed enzymatic activities leukocyte subtypes CD73 PAP knockouts generated CD73/PAP double knockout mice elucidate contribution immunological parameters. Enzymatic assays confirmed ability recombinant human hydrolyze [(3)H]AMP, although at much lower rate than CD73. Nevertheless, 5'-nucleotidase/AMPase splenocytes lymphocytes from PAP(-/-) tended be wild-type controls, suggesting potential PAP, along with into lymphoid metabolism ex vivo. Single had decreased number CD4(+)/CD25(+)/FoxP3 (+) regulatory T cells thymus exhibited reduced percentages CD4(+) spleen, lymph nodes thymus, CD8(+) blood. These findings suggest has a synergistic together by contributing balance subpopulations especially thymus.
