摘要: We have created missense mutations in the indirect flight muscle (IFM)-specific Act88F actin gene of Drosophila melanogaster by random vitro mutagenesis. Following P element-mediated transformation into wild-type flies and subsequent transfer inserts null strains, effects mutants on structure function IFMs were examined. All antimorphic for ability. E316K G368E formed with only relatively small defects whilst others produced large amounts disruption. E334K filaments but lacked Z discs. V339I no did not accumulate actin. E364K G366D both had stable form myofibrils. Using an polymerisation assay we found significant ability mutant actins to polymerise. also caused a strong induction heat shock protein (hsp) synthesis at normal temperatures accumulated hsp22 which, together actin, was resistant detergent extraction. Both weak hsp synthesis. discuss how stability, different affects myofibril assembly function, hsps.
