摘要: Guzhi Zengsheng Zhitongwan (GZZSZTW) is an effective formula of traditional Chinese herbal medicine and has been widely applied in the treatment joint diseases for many years. The aim this study was to dissect molecular targets signaling pathways based on analysis serum proteomics. herbs GZZSZTW were immersed 5 l distilled water boiled with reflux extraction method. extract filtered, concentrated freeze-dried. chemical profile determined by high-performance lipid chromatography (HPLC). 7-week old Sprague-Dawley (SD) rats groups received oral administration at doses 0.8, 1.05, 1.3 g/kg per day blank group fed drinking water. Serum samples collected from jugular veins. Primary chondrocyte viability evaluated CCK-8 assay. A full spectrum investigated isobaric tags relative absolute quantitation (iTRAQ) a systematic bioinformatics accompanied parallel reaction monitoring (PRM) siRNA validation. regulated series functional proteins responsible cartilage development, growth repair. Functional classification indicated that these mainly involved process cell surface dynamics. Pathway mapped into several signalling chondrogenesis, proliferation differentiation, repair, including hippo pathway, cGMP-PKG cycle calcium pathway. Protein–protein interaction knockdown assay identified network consisting TGFB1, RHO GTPases, ILK, FLNA, LYN, DHX15, PKM, RAB15, RAB1B GIPC1. Our results suggest effects treating might be achieved through TGFB1/RHO coupled other Therefore, present greatly expanded our knowledge provided scientific support underlying therapeutic mechanisms diseases. It also possible alternative strategies prevention using formulas.
