摘要: The epidermal growth factor receptor (EGFR) is a tyrosine kinase involved in various cellular processes including cell differentiation, proliferation, and migration. Despite many years of intensive research its mode activation still not fully understood, partly because dimerization membrane localization are under discussion. Here we use Single Wavelength excitation Fluorescence Cross-Correlation Spectroscopy (SW-FCCS) Imaging Correlation (Imaging FCS), to investigate membranes. The fraction EGFR preformed dimers, i.e. the absence ligands, has been determined conditions with very different results, ranging from exclusive monomeric states distributions between monomers dimers possibly higher oligomers. reason for these discrepancies unclear. We therefore measure on lines (CHO-K1, Cos7, HEK293) at locations (basal apical membrane) room physiological temperature determine whether sensitive conditions. observe line specific sensitivity that can change detected dimer by more than 2, hinting an explanation why experiments provide answers question dimers. Furthermore, FCS within interaction cytoskeleton. Our study shows interacting cholesterol dependent independent domains motion influenced Overall measurements complex picture organization dynamics, which might play essential roles function signaling.
