摘要: To get a better insight into the relationship between cell wall integrity and pathogenicity of fungus Botrytis cinerea, we have constructed chitin synthase mutants. A 620 bp class I gene fragment (Bcchs1) obtained by PCR amplification was used to disrupt corresponding in genome. Disruption Bcchs1 occurred at frequency 8%. Nine independent mutants were mutant phenotype compared that transformants which not disrupted. These disruption dramatically reduced their vitro Mg2+, Mn2+, Co2+-dependent activity. Chitin content 30%, indicating Bcchs1p contributes substantially composition. Enzymatic degradation cocktail glucanases revealed weakening mutant. transcribed constant level during vegetative exponential growth, suggesting it necessary throughout hyphal development. growth identical undisrupted control transformant however, exhibited on vine leaves. It can be assumed leads might slow down planta fungal progression.
