Study of the binding of nuclear proteins from Plasmodium berghei strains with different chloroquine sensitivity to oligonucleotides corresponding to regulatory elements of the multidrug resistance gene

摘要: Using electrophoretic mobility shift assay the proteins, specifically bound to double-stranded oligonucleotides corresponding binding sites of transcription factors AP1 family, NF-IL6 and SP1 involved in up-regulation human multidrug resistance (mdr1) gene also proteins oligonucleotide element responsive for stimulation by serum (SRE) have been found nuclear extracts rodent malaria parasite Plasmodium berghei (P. berghei). P. strains exhibiting different chloroquine sensitivity are characterized patterns protein used. Mutations consensus sequences AP1, NF-IL6, SRE caused impairments some proteins; this suggests existence containing DNA domains, which share similarity with domains SRF1 members family. The results obtained suggest profound alterations regulatory apparatus plasmodium during its selection resistance.
