Rapid visualization of hydrogen positions in neutron protein crystallography structures

摘要: Neutron crystallography is a powerful technique to visualize experimentally the position of light atoms, including hydrogen and its isotope deuterium. Over last several years, structural biologists have shown an increasing interest for as it uniquely complements X-ray crystallographic data by revealing hydrogen/deuterium atoms in macromolecules. With this regained interest, access macromolecule neutron beam lines becoming limiting step. In report we show that rapid collection could be valuable alternative long time when appropriate. Comparison perdeuterated Rubredoxin structures refined against sets collected over hours up five days shows just 14 sufficient provide 262 positions without ambiguity.
