摘要: Abstract Adult hematopoiesis requires tightly regulated cell–cell interactions between hematopoietic cells and the bone marrow stromal microenvironment. We addressed question if ectodomain sheddase ADAM10 is essential to regulate adult hematopoiesis. Induced deletion in resulted morphological histological abnormalities that resemble an unclassified myeloproliferative disorder (MPD). The MPD was characterized by expansion of granulocytic subpopulations their infiltration peripheral tissues, development hepatosplenomegaly with extramedullary erythropoiesis, lymphnodepathy death mice around 20 weeks after induction. expression analysis during different stages revealed non-targeted repopulated immune system ADAM10-deficient mice. Examination a myeloid- or epidermis-specific transplantation (BMT) experiments indicated can be triggered non-cell autonomous effects. found plasma levels clinical markers for such as G-CSF, TIMP-1 IL-16 were significantly elevated Our findings indicate controlled needed balance cell-fate decisions
