摘要: Abstract The function of prothymosin alpha has been investigated by using four different antisense oligodeoxyribonucleotides directed at selected regions its mRNA. In every case, when synchronized human myeloma cells were released from stationary phase incubation in fresh medium containing oligomers, cell division was prevented or inhibited; sense oligomers and random had no effect. A detailed analysis populations indicated that sense-treated untreated divided approximately 17 hr after growth initiation, whereas incubated with oligomer 183, a 16-mer targeted 5 bases downstream the initiation codon, entered mitosis one late. The failure to divide correlated directly deficit continued presence intact intracellular over period least 24 hr. Because effect either on timing induction mRNA upon stimulation levels seen throughout cycle, we concluded DNA caused specific hybrid arrest translation. Our data suggest is required for division. However, there evidence regulates mitosis.
