摘要: Characterization and quantification of carotenoid compound is complicated, costly timeconsuming. The accuracy reliability the data depend solely on standard to High Performance Liquid Chromatography (HPLC) analysis but major constraint acquire maintain pure standards. Carotenoid standards are commercially available they expensive prone isomerization and oxidation. Thus, purpose this study establish an analytical method for isolating β-carotene by using open column chromatography (OCC) from pumpkin (Cucurbita moschata) be used as one carotenoid standards determination total individual carotenoid. Pumpkin with orange flesh has been chosen due non-seasonal nature its availability all year-round. This demonstrated that purity of β-carotene standard; determined HPLC was ranged 92.21 97.95%. The curves five different concentrations extract pumpkins in triplicate were constructed plotting peak area against concentration. coefficient correlation 0.9936. Therefore, established that pumpkin can a reliable source beta-carotene it cheap commonly throughout the year.
