Enhanced production of single copy backbone-free transgenic plants in multiple crop species using binary vectors with a pRi replication origin in Agrobacterium tumefaciens.

摘要: Single transgene copy, vector backbone-free transgenic crop plants are highly desired for functional genomics and many biotechnological applications. We demonstrate that binary vectors use a replication origin derived from the Ri plasmid of Agrobacterium rhizogenes (oriRi) increase frequency single in tumefaciens mediated transformation soybean, canola, corn, compared to RK2-derived (RK2 oriV). In large scale soybean experiments, was nearly doubled two versions oriRi RK2 oriV control vector. canola produced more than did corn copy also significantly increased when using vector, although dropped. These results, experiments three crops, indicate advantage over production Agrobacterium-mediated transformation.
