摘要: Importance Risk factors associated with the development of neovascular age-related macular degeneration (AMD) have been identified. However, population size and methods to integrate imaging, genetic, demographic conversion AMD are limited, specifically when treatment is administered in 1 eye. Objective To determine from nonneovascular fellow eyes. Design, Setting, Participants This post hoc secondary analysis 24-month phase 3 multicenter, double-masked, active treatment-controlled HARBOR trial included 686 eyes at baseline. Imaging features describing presence, number, extent, density, relative reflectivity drusen were automatically extracted spectral-domain optical coherence tomography scans. Genetic 34 single-nucleotide polymorphisms. Least absolute shrinkage selection operator regression was performed narrow imaging features. Survival Cox proportional hazards association selected genetic AMD. Data collected November 2016 through October 2017 analyzed 2018. Exposure Nonneovascular Main Outcomes Measures Features Hazard ratios (HRs) their 95% CIs calculated. Results Among (406 [59.2%] women; mean [SD] age, 78.12 [8.28] years), 154 (22.4%) converted Female sex significantly (HR, 1.57; CI, 1.11-2.20; P  = .009). After controlling for effects, area within mm fovea (HR, 1.45; 1.24-1.69; HR 1-SD increase, 1.36 [95% 1.20-1.54]) 3.97; 1.11-14.18; 1.32 1.02-1.71]) In addition, variant (rs61941274) found be Conclusions Relevance Two (total en face restricted a circular reflectivity) ( ACAD10 locus) Drusen characteristics may despite Trial Registration ClinicalTrials.gov identifier:NCT00891735
