Antisense flavonol synthase alters copigmentation and flower color in lisianthus

摘要: In order to generate new flower colors in lisianthus, a flavonolsynthase (FLS) coding sequence was isolated from lisianthus (Eustomagrandiflorum Grise.) using petunia homologue (pCGP481) as probe.The endogenous FLS mRNA transcript occurs early petal development,concomitant with accumulation of flavonols bud tissue, and ceases at onsetofanthocyanin pigment production. Southern DNA analysis indicated memberof multigene family lisianthus. Transgenic plants purple floweredlisianthus line expressing antisense under the control CaMV35Spromoter produced flowers more red color (magenta) than originaluntransformed plant. The transgenic also showed novel pigmentation(cyanidin) stage buds well deeply colored pistils, andaccumulated dihydroflavonols expense flavonols. Further, ofthe progeny crosses between primary transformants pigmentedpurple variety (Wakamurasagi) this phenotype be stablyinherited. Field trial assessment variationinthe although variation flowercolor within range normally seen commercial cultivars grown undercommercial conditions.
