Establishment and application of an accurate identification method for fragrant soybeans

摘要: Abstract In order to screen the aroma characteristics of soybean, a new method was established which can quickly quantify content 2-acetyl-1pyrroline (2-AP), an important compound related soybean aroma, using gas chromatography-mass spectrometry (GC-MS). Based on peak profile, total area and retention time as test indexes, accurate identification for fragrant soybeans established. The optimum parameters protocol consisted column temperature 70°C, sample injector 180°C, extraction alcohol 1 mL, NaCl 0.1 g, ultrasonication 10 min, h, were by orthogonal single factors three with four levels (L9 (3)4). 2-AP leaves had significant correlations seeds, easier measure. simple easy carry out, consumed only small amounts reagents, provided reliable results good reproducibility. A 101 genotypes from different geographical sources analyzed this protocol. showed that average 0.29 mg L−1, ranging 0.094 1.816 genetic diversity index 0.54. Among all genotypes-tested, they classified into grades, including seven elite identified “grade one soybeans”, Zhonglong 608, Heinong 88, Ha13-2958, Hongmiandou, 82, Huangmaodou, Jiyu 21. These provide both technique several gene discovery quality breeding in soybean.
