摘要: A new toxin of Enterobacter cloacae able to lyse erythrocytes and leukocytes was found. Purification the performed by salt precipitation, gel filtration, ion exchange HPLC in C8 column. SDS-PAGE electrophoresis showed more than one bank corresponding leukotoxin form polymers aggregate like some pore-forming cytotoxins (RTX). In culture supernatant 1 HU/ml (hemolytic unit) 1.5 LU/ml (leukotoxic unit); after purification it reached 15 20 LU/ml. The ratio between HU percentage red cells affected lytic capacity. E. stimulated oxidative metabolism neutrophils, but over 50 μg toxin/ml stimulus ceased as shown NBT assay due cell death. Chemiluminescence evidenced an increase superoxide anion generation, excess interfered with this stimulus, previously observed HlyA Escherichia coli toxin. Cross-reaction found immunoblotting HlyA. presented higher amounts proline, valine, aspartic glutamic acids similar prescence a glycine-rich DNA sequence effect calcium on activity. did not cross-react hemolysin HmpA Proteus.
