摘要: Osteopontin protects endothelial cells from apoptosis induced by growth factor withdrawal. This interaction is mediated the alpha(v)beta(3) integrin and NF-kappaB-dependent (Scatena, M., Almeida, Chaisson, M. L., Fausto, N., Nicosia, R. F., Giachelli, C. (1998) J. Cell Biol. 141, 1083-1093). In present study we used differential cloning to identify osteopontin-induced, genes in cells. One of identified this screen was osteoprotegerin, a member tumor necrosis receptor superfamily. By Northern Western blot analysis, osteoprotegerin mRNA protein levels were very low plated on non-integrin cell attachment factor, poly-d-lysine. contrast, 5-7-fold following ligation osteopontin. Osteoprotegerin induction osteopontin time-dependent observed as early 3 h treatment. NF-kappaB inactivation achieved over expression an IkappaB super repressor completely inhibited Finally, purified protected with inactive deprivation. These data suggest that alpha(v)beta(3)-mediated survival depends indicate new function for
