Effect of acute betaine administration on hepatic metabolism of S-amino acids in rats and mice.

摘要: Abstract Alterations of hepatic glutathione level by betaine were observed previously. In this study effects administration (1000 mg/kg, i.p.) on S -amino acid metabolism in rats and mice investigated. Hepatic decreased rapidly followed marked elevation 24 hr. Concentrations -adenosylmethionine, -adenosylhomocysteine, methionine increased whereas cystathionine significantly, suggesting that homocysteine generated the cycle is preferentially remethylated to rather than being utilized for synthesis cysteine. cysteine concentration declined immediately, but plasma increased. Effect uptake was estimated from difference major blood vessels connected liver. Cysteine either portal vein or abdominal aorta not altered, however, a significant increase noted vein, indicating treatment. Activities glutamate ligase, β-synthase, γ-lyase elevated Pretreatment with propargylglycine, an irreversible inhibitor γ-lyase, did abolish betaine-induced reduction 4 hr, at t =24 hr blocked completely. conclusion present results indicate induces time-dependent changes acids. Betaine enhances metabolic reactions cycle, inhibits uptake, leading decrease supply synthesis. Reduction subsequently reversed due induction ligase activity.
