摘要: Nisin Z, a natural nisin variant, was recently isolated from Lactococcus lactis subspecies NIZO 22186. The gene for this lantibiotic, designated nisZ, has been cloned, and its nucleotide sequence found to be identical that of the precursor with exception single mutation resulting in substitution Asn-27 His-27 mature polypeptide (J. W. M. Mulders, I. J. Boerrigter, H. S. Rollema, R. Siezen, de Vos, Eur. Biochem. 201:581-584, 1991). A K+ electrode used investigate effect various environmental parameters on action Z against Listeria monocytogenes. Addition resulted immediate loss cell K+, depolarization cytoplasmic membrane, inhibition respiratory activity, hydrolysis partial efflux cellular ATP. optimal at pH 6.0 significantly reduced by di- trivalent cations. lanthanide gadolinium (Gd3+) an efficient inhibitor prevented activity completely concentration 0.2 mM. Z-induced low temperatures, presumably as result increased ordering lipid hydrocarbon chains membrane. In cells grown 30°C, below 7°C, whereas 4°C able induce leakage temperature.
