摘要: Studies have shown that human interferon inducible transmembrane protein (hIFITMs) family proteins broad-spectrum antiviral capabilities. Preliminary studies in our laboratory tentatively proved hIFITMs the effect of inhibiting influenza viruses. In order to further study its mechanism and role occurrence development A, relevant been carried out. Fluorescence quantitative polymerase chain reaction (PCR) detection technology was used observe hIFITM3 on replication A virus (IVA) interaction with hABHD16A. HEK293 cells, overexpression significantly inhibited IVA at 24 h, 48 h, 72 h; yeast two-hybrid experiment interacts hABHD16A; laser confocal microscopy observations showed hABHD16A colocalized cell membrane area; expression level inflammation-related factors cells overexpressing or detected by fluorescence PCR, results mRNA levels interleukin- (IL-) 1β, IL-6, IL-10, tumor necrosis factor- (TNF-) α, cyclooxygenase 2 (COX2) were increased. But when hIFITM3/hABHD16A coexpressed, these cytokines reduced except COX2. When infected coexpressing hIFITM3/hABHD16A, inflammatory decreased compared control group, indicating can play an important regulating inflammation balance. This confirmed has replication. Furthermore, it found hABHD16A, following which better inhibit response caused disease process.
