Effects of concentration variation on the physical properties of alginate-based substrates and cell behavior in culture.

摘要: Nowadays alginate capsules exhibit good biocompatibility and high permeability for nutrients metabolic wastes making them appealing biomaterial therapeutic cell encapsulation. Further study of the characteristics beads which are highly dependent on various environmental conditions to create an optimum microenvironment cells is also critical. Thus, in this study, effect concentration variation physical properties alginate-based entrapped-cells behavior was analyzed. Results showed that increase Ca ions brought about decrease average diameter, prolongation dissolution time, reduction swelling, a rise crosslinking extent shrinkage capsules; while raising sodium had opposite diameter shrinkage. Moreover, addition gelatin enhanced penetration swelling slowed down capsules. And MC3T3-E1 enclosed particles calcium chloride, 2.5%, 2.0% 0.5% (w/v %) preferable abilities proliferation higher expression alkaline phosphatase. Overall, ability tailor system support vitro growth might have significance future use other types regenerative medicine.
