Spatial and temporal regulation of Wnt/β-catenin signaling is essential for development of the retinal pigment epithelium

摘要: Wnt/β-catenin signaling is highly active in the dorsal retinal pigment epithelium (RPE) during eye development. To study role of RPE development we used a conditional Cre/loxP system mice to inactivate or ectopically activate RPE. Inactivation results transdifferentiation neural retina (NR) as documented by downregulation RPE-specific markers Mitf and Otx2 ectopic expression NR-specific Chx10 Rx, respectively. In contrast, activation disruption patterning, indicating that precise spatial temporal regulation required for normal Using chromatin immunoprecipitation (ChIP) reporter gene assays provide evidence isoform Mitf, Mitf-H, are direct transcriptional targets signaling.
