摘要: Both D- and L-serine in rat brain microdialysis sample were simultaneously determined by pre-column fluorescence derivatization with 4-fluoro-7-nitro-2,1,3-benzoxadiazole (NBD-F), separation of the derivatives on ODS column, TSKgel ODS-80TsQA, followed Pirkle type chiral columns, Sumichiral OA-2500 (S), which gave a sufficient enantiomeric NBD-D-serine NBD-L-serine, fluorimetric detection at wavelength 540 nm an excitation 470 nm. The peaks NBD-L-serine clearly found, validation study showed satisfactory results; precision accuracy within 5.14 109%, respectively. Using proposed HPLC method, time-course profile D-serine concentration prefrontal cortex following intraperitoneal administration was investigated. As consequence, appeared to be rapidly distributed brain, then decreased gradually time extracellular fluid cortex. method will useful for vivo studies D-serine, acts as coagonist N-methyl-D-aspartate receptor, brain.
