Proliferation and death of cultured fetal neocortical neurons: effects of ethanol on the dynamics of cell growth.

摘要: Neuronal number in the mature CNS is determined by balance of cell proliferation and death. The effects ethanol on death were examined primary cultures neocortical neurons derived from 16-day-old rat fetuses. cells treated with (0 or 400 mg/dl) for (1) immunohistochemical identity, (2) cycle kinetics using a cumulative bromodeoxyuridine labeling technique, (3) viable via trypan blue assay, (4) incidence terminal deoxy-nucleotidyl transferase-mediated dUTP nick end (TUNEL) caspase 3 immunhistochemistry. After two days culture, most (>85%) expressed neuron-specific antigen(s) whether not was added to culture medium. Ethanol affected cultured cells, e.g., length greater ethanol-treated than controls. blue-negative (viable) profoundly decreased exposure. This decrease accompanied increases frequencies TUNEL- 3-positive exhibiting nuclear condensations. Thus, decreases vitro slowing increasing expression indices untreated consistent single (apoptotic) pathway death, rather simultaneous apoptotic necrotic modes Furthermore, it appears that initiates an among cortical neurons.
