摘要: Transforming growth factor β (TGFβ) is a homodimeric polypeptide that enhances the aromatase activity in rat granulosa cells. So far, effect of TGFβ on cells has been primarily studied estrogen-primed immature rats. Our study designed to investigate TGFβx steroidogenesis cultured mature Mature were obtained from ovarian follicles adult rats have demonstrated 3 consecutive 4-day estrous cycles. The treated by subcutaneous injections 20-IU pregnant mare serum gonadotropin (PMSG) at 2000h day estrus followed 15-IU PMSG 0800 and next day. In each experiment, total 10 ovaries collected for cell preparation 40h after last injections. average viability was about 80%. culture wells precoated with Dulbecco’s Modified Eagle Medium/Ham’s F-12 nutrient mixture (50:50) (DME/F-12) supplemented 10% fetal calf (FCS) under 5% CO2, 95% air 37°C 24h, media removed washing times serum-free DME/F-12. Then, well seeded approximately 15,000 live cells/mL. DME/F-12 1 μg insulin, 1-IU thrombin 10-ng low-density lipoprotein per mL (DME/F-12/S) 48h.
