Effects of perfluorooctane sulfonate on the conformation and activity of bovine serum albumin

摘要: Perfluorooctane sulfonate (PFOS) is among the most prominent contaminates in human serum and has been reported to possess potential toxicity body. In this study, effects of PFOS on conformation activity bovine albumin (BSA) were investigated vitro. The results indicated that binding interaction with BSA destroyed tertiary secondary structures protein loss α-helix structure increasing hydrophobic microenvironment Trp or Tyr residues. During thermal denaturation protein, increases stability BSA. proportion decreased concentration residues becomes more hydrophobic. from molecular modeling had not only one possible site bind by polar interaction, hydrogen bonds forces. addition, relative activities increase concentration. Such presence sites located subdomain IIIA, which good agreement fluorescence spectroscopic experiments results. This study offers a comprehensive picture interactions provides insights into toxicological effect perfluoroalkylated substances.
