Localization of Fructan Metabolism in the Vacuoles Isolated from Protoplasts of Jerusalem Artichoke Tubers (Helianthus tuberosus L.)

摘要: Summary Protoplasts were prepared from tubers of Helianthus tuberosus in the developing and resting stage development. Vacuoles isolated protoplasts purified by sedimentation through a density gradient glycine betaine. All fructan (inulin) with DP ⩾ 3 (i.e. trisaccharide isokestose larger polymers fructose) was found to be located exclusively vacuoles whilst sucrose, glucose, fructose only partially there. The also sole cell compartments containing synthesizing enzyme activities (sucrose-sucrose-fructosyl-transferase fructan-fructan-fructosyl-transferase) degrading activity (fructan exohydrolase), depending on development tubers. vacuole is therefore proposed organelle for storage metabolism.
