摘要: Lipid peroxidation due to oxidative stress is accelerated under hyperglycemic conditions such as diabetes mellitus. The effect of 4-hydroxy-2-nonenal (HNE) and other lipid products on the ability isolated rat pancreatic islets secrete insulin was examined in this study. HNE concentration- time-dependently deteriorated glucose-induced secretion: secretion decreased by 50% when measured after incubation with 100 microM for 1 h. Other products, e.g. 2-hexenal 2-butenal, also inhibited secretion. at lowered alpha-ketoisocaproate-induced secretion, whereas leucine-induced stimulated. Insulin induced 10 mM glyceraldehyde slightly HNE. On hand, severely 2.8 glucose. Glucose utilization glucose oxidation were significantly treated amounts fructose 1,6-bisphosphate dihydroxyacetone phosphate treatment HNE, amount 6-phosphate increased. Our study indicates that impair probably through affecting both glycolytic pathway citric acid cycle.
