Involvement of TNF-Like Weak Inducer of Apoptosis in the Pathogenesis of Collagen-Induced Arthritis

摘要: TNF-like weak inducer of apoptosis (TWEAK) is a type II membrane protein belonging to the TNF family that regulates apoptotic cell death, cellular proliferation, angiogenesis, and inflammation. However, role TWEAK in pathogenesis rheumatoid arthritis (RA) remains unclear. In this study, we have investigated effect neutralizing anti-TWEAK mAb on development collagen-induced (CIA), well-established murine model RA. Administration significantly ameliorated paw swelling, synovial hyperplasia, infiltration inflammatory cells. The levels proinflammatory chemokines such as MCP-1 MIP-2 serum knee joints were reduced by treatment. Consistently, recombinant enhanced proliferation production cells from CIA mice vitro. Histological examination also revealed treatment with suppressed small vessels tissues. These results indicated anti-inflammatory antiangiogenic effects blockade CIA, which may be beneficial for
