摘要: The standard products of V(D)J recombination immunoglobulin and T cell receptor genes are two kinds DNA junction, a coding joint signal joint. TCR delta V-D D-D joints in adult mouse thymocytes were sequenced following PCR amplification. We observed differential nucleotide insertions at the V delta-D joints, depending on D gene usage developmental stage. Nucleotide 1 less frequent for 4, 5 preferentially utilized than 3, 6 genes, infrequently rearranged to 1. In addition unexpectedly, novel nonstandard products, "replacement joints" substituted downstream by sequence also found. However, no 2-associated replacement other 1-D 2 observed. mutation beta affected frequency inhibited formation Recombination mechanism generating possible role up-regulation rearrangements discussed.
