Quantitative immunoferritin microscopy of Fya, Fyb, Jka, U, and Dib antigen site numbers on human red cells

摘要: The Fya, Fyb, Jka, U, and Dib antigen site numbers ultrastructural distribution patterns on the human erythrocyte membrane were determined using quantitative immunoferritin microscopy. For homozygous antigen-positive red cells, average number of determinants per cell was about 14,000 for 17,000 Fya 19,000 Dib, 23,000 U antigen, assuming that equilibrium binding observed represented 80% saturation accessible sites. this group antigens less than Rh antigens, but considerably more Kell Cellano antigens. technique used capable demonstrating a twofold difference in density between heterozygous Fy (a+) cells. More 85% Fyb sites lost following pretreatment cells with papain, consistent serologic lability proteolysis. ferritin conjugate staining antibody-sensitized ghost membranes similar all five studied showed random, clustered pattern. Although estimates are valid, remarkable similarity pattern diverse as well other considerations, suggest findings ghose probably do not reflect faithfully arrangement intact membrane.
