摘要: Protein folding in the living cell begins cotranslationally. To analyze how it is influenced by ribosome and translocon complex during translocation into endoplasmic reticulum, we expressed a mutant influenza hemagglutinin (a type I membrane glycoprotein) with C-terminal extension. Analysis of nascent chains two-dimensional SDS-PAGE showed that attachment as such had little effect on ectodomain or trimer assembly. However, long were bound inside complex, formation disulfides was partially suppressed, trimerization inhibited, protein protected against aggregation.
