摘要: Electron-microscopic studies with peroxidase cytochemistry have shown that primary (azurophilic) granules in human neutrophils are synthesized promyelocytes, while secondary (specific) formed myelocytes. However, these were limited by the lack of specific markers for and inability to make direct comparisons electron-microscopic morphology light-microscopic appearance same cell. Thus cannot be identified reliably relevance findings interpretation clinical bone marrow specimens evaluated. To circumvent problems, we developed a method permit immunofluorescent demonstration granule cells stained Romanowsky agents. Normal May-Gruenwald-Giemsa photographed. The slides decolorized buffered glycerine saline 24 hr then fluorescein- rhodamine-conjugated monospecific antisera granulocyte myeloperoxidase, cathepsin G, elastas, lysozyme, lactoferrin. located examined conjugate binding. Double stains rhodamine-labeled offered additional advantage simultaneous identification granules.
