摘要: Transcription factor NF-κB regulates the physiological response to a variety of stimuli. The pathway has served as paradigm for analyzing impact covalent protein modifier ubiquitin on signal transduction. discovery in early 1990s that degradation cytosolic inhibitors (IκBs) is mediated by proteasome system (UPS) was first example direct involvement ubiquitination cellular signaling. By now it become clear role extends far beyond triggering IκB destruction. kinase (IKK) complex key regulator NF-κB. Attachment chains IKK and further upstream components drives signaling pathways promoting clustering network. Whereas conjugation serves positive function pathway, deconjugation acts negative regulatory feedback mechanism critically involved balancing strength duration response. Moreover, inactivation deconjugating enzymes can cause sustained activity under pathological conditions like chronic inflammation or cancer.
