Affinity purification of a major Alternaria allergen using a monoclonal antibody.

摘要: An extract of Alternaria (ALT) was passed through an affinity column containing a monoclonal antibody directed to ALT. After washing the column, single glycoprotein eluted using 0.1 M glycine (pH 3.0). This accounted for 13% dry weight our ALT preparation and had carbohydrate protein ratio 0.35 as compared with 3.2 whole extract. Its molecular 70 kD by SDS-PAGE isoelectric point 3.5 IEF. Though individual sensitivities varied, 14/16 patients skin reactive were also this glycoprotein. Quantitative tests extracts adjusted same per volume showed that it required about four times greater concentration purified give wheal size equal Alternaria.
