摘要: Eight different amber suppressor tRNA (suptRNA) mutations in the nematode Caenorhabditis elegans have been isolated; all are derived from members of tRNA(Trp) gene family (K. Kondo, B. Makovec, R. H. Waterston, and J. Hodgkin, Mol. Biol. 215:7-19, 1990). Genetic assays activity suggested that individual genes were differentially expressed, probably a tissue- or developmental stage-specific manner. We now examined expression representative this both vitro, using transcription embryonic cell extracts, vivo, by assaying suppression an amber-mutated lacZ reporter animals carrying suptRNA mutations. Individual wild-type their amber-suppressing counterparts appear to be transcribed processed identically suggesting behavior suptRNAs should reflect expression. The levels closely parallel extent genetic vivo. results suggest differential is most likely at transcriptional rather than posttranscriptional level 5' flanking sequences play role vivo as well. Using lacZ(Am) more direct assay types, we again observed which correlates with vitro results. This provides model system extensively study basis for family.
