摘要: The identification of novel cell lines which combine the most important properties mucosal membranes in terms drug absorption, transmembrane transport and mucus secretion can help to establish improved meaningful test systems for pharmacological infectiological studies. We have established a secreting tumor line (Cx-03) derived from female patient who underwent radical hysterectomy after diagnosis large malignant carcino sarcoma (Muellerian mixed tumor). Via xenotransplantation SCID beige mice, recultivation subcloning stable was primary cells. Human origin novelty determined by karyotype analysis STR fingerprint. During growth cells produce considerable amounts PAS positive viscoelastic mucus. Immunostaining revealed expression mucins mucin modifier CLCA1. demonstrate initial electrophysiological experiments that confluent, polarized monolayers Cx-03 are formed (on PCF-filter supports) exhibit electrical resistance (> 600 Ω cm2). Confluent express barrier-forming tight junction proteins claudin-1 -4 co-localize with zonula occludens protein-1 (ZO-1) at cell-cell contacts. Mucus is rare property among mammalian lines. In combination its ability form might contribute as based model barrier
