Structure and expression of the ABF1-regulated ribosomal protein S33 gene in Kluyveromyces.

摘要: The abundant multifunctional protein ABF1 of Saccharomyces cerevisiae binds to the upstream region several genes, including some ribosomal genes like one encoding S33. Deletion ABF1-binding sequence lowers transcription these three- more than ten-fold. We have isolated S33 two related yeast species, Kluyveromyces lactis and marxianus. Comparison nucleotide sequences with their counterpart from S. shows a strong similarity covering whole coding regions. In contrast, little or no is found in 5'-flanking regions three genes. Also trailer differ considerably both length species another. An site present gene K. Retardation analyses showed that this able bind cells molecular mass somewhat lower ABF1. Functional analyses, using beta-glucuronidase reporter system, indeed involved activation marxianus cells. A ABF1-gene-specific probe only weak hybridization DNA Northern blots did not show signal. These results indicate contain functionally but structurally dissimilar ABF1-type proteins.
