摘要: In view of the limited stability isourea bond, formed in ligand coupling to CNBractivated polysaccharides, an alternative this current activation method has been developed. 2,4,6-Trifluoro-5-chloropyrimidine (FCP), known as a reactive group dyes, was used activate Sepharose. Under appropriate conditions thermally stable product with unimpaired beaded structure obtained, which toward amines and mercaptans. Coupling hexamethylenediamine, aniline, ethanethiol, respectively, yielded incorporation 0.2-2.7, 0.9-1.7, 1.1 mmol ligand/g dry agarose. The immobilized ligands based on FCP-Sepharose between pH 4 8 about 200 times higher compared products originating from CNBr-Sepharose; leakage only 0.5 x 10p-3%/h. possibility obtaining high degree substitution is further advantage FCP activation. addition, FCP-activated Sepharose can be stored wet state at 4°C without substantial decrease capacity. analogs 2,4,5,6-tetrachloro- 2,4,5,6-tetrafluoropyrimidine, other polymers (cellulose, Sephadex, aminomethylpolystyrene) appeared applicable also.
