Cellular factors required for multiple stages of SV40 DNA replication in vitro.

摘要: Plasmids containing the SV40 origin replicate in presence of T antigen and a cell free extract derived from human 293 cells. Upon fractionation this extract, two essential replication factors have been identified. One these is multi-subunit DNA binding protein polypeptides 70,000, 34,000 11,000 daltons which may function as eukaryotic single strand (SSB). The other partially purified fraction required with for first stage replication, formation pre-synthesis complex at origin. These results, others, define multiple stages vitro are analogous to Escherichia coli phage lambda reflect similar events cellular chromosomes.
