摘要: Na+/H+ exchanger (NHE) activity is exquisitely dependent on the intra- and extracellular concentrations of Na+ H+. In addition, Cl- ions have been suggested to modulate NHE activity, but little known about underlying mechanism, sensitivity individual isoforms has not established. To explore their sensitivity, types 1, 2, 3 exchangers (NHE1, NHE2, NHE3) were heterologously expressed in antiport-deficient cells. Bilateral replacement with nitrate or thiocyanate inhibited all isoforms. depletion did affect cell volume cellular ATP content, which could indirectly altered activity. The number plasmalemmal was unaffected by removal, implying that inhibition due a decrease intrinsic exchangers. Analysis truncated mutants NHE1 revealed anion resides, at least part, COOH-terminal domain exchanger.
