Molecular characterization, tissue-specific expression, and RNA knockdown of the putative heat shock cognate 70 protein from Rhipicephalus haemaphysaloides

摘要: Heat shock cognate 70-kDa protein (RH-Hsc70) was identified from a cDNA library synthesized the sialotranscriptomes of unfed and fed Rhipicephalus haemaphysaloides. The RH-Hsc70 open reading frame is 1950 bp long encodes that 649 amino acids in length, with predicted molecular weight 71.1 kDa theoretical pI 5.43. exhibits 98% acid identity Hsc70 Haemaphysalis flava 83% arthropods mammals. mainly expressed nymphs adult ticks, not larvae. Real-time quantitative PCR analysis indicated mRNA expression induced by blood feeding ticks. In addition, gene higher ovaries ticks than midguts, salivary glands, fat bodies or knockdown inhibited tick feeding, significantly decreased engorgement rate, increased death rate. These data illustrate importance aging, which makes it promising candidate for development anti-tick vaccines.
